In vitro regeneration study in lablab bean and dolichos bean (Lablab purpureus (L). Sweet) Genotypes

Abstract: The current investigation was carried out to study in vitro regeneration potential in Lablab bean genotypes and Dolichos bean Cv. (Konkan Bhushan). In surface sterilization, treatment with 70% alcohol for 5 minute followed by HgCl2 (0.1%) for 10 minutes was found the most effective in establishing aseptic cultures (97.22%). Mature embryo axis with single cotyledon (MEASC) explant on medium MS+ 6 mg/l BAP+ 0.5 mg/l NAA responded well for shoot induction frequency, multiple shoot induction in Dolichos bean genotype ‘Konkan Bhushan’. The medium MS+ 0.5mg/l BAP+ 0.2 mg/l GA3 was found better for elongation of in vitro regenerated shoots. The genotype ‘Konkan Bhushan’ rooted also earliest in 9.30 days on medium MS+ 1.0 mg/l IBA+ 2% sucrose with highest root induction frequency (79.51%) and maximum roots per shoot (5.04). During hardening, the potting mixture of soil, vermiculite and sand in 1:1:1 showed the highest survival rate.
