Abstract:
The present study was aimed to develop and validate stability indicating studies
Andrographis paniculata by HPTLC. The separation was attained by using precoated TLC plates with silica gel 60F
254. The mobile phase used was chloroform-toluene-methanol (7:1.5:1.5, v/v/v). Densitometric analysis was carried out at 254 nm. The linear regression analysis data obtained for the calibration plots displayed good linear relationship with respect to peak height and peak area, in the concentration range 100-500 ng/spot. The limits of detection (LOD) and quantification (LOQ) were found to be 0.52 ng/spot and 1.59 ng/spot, respectively. Stability indicating studies exhibited the well resolved peak of the standard andrographolide and a complete degradation in case of extract of
Andrographis paniculata. The present study describes the degradation of standard andrographolide and the extract of
Andrographis paniculata under different stress conditions according to the ICH guidelines i.e. acid, alkali, dry heat, oxidation and photolytic degradation.
